The present invention relates to a kit for detecting a target substance, comprising two segments divided from G-quadruplex. The present invention also relates to a method for determining the presence of a target substance comprising a step of treating the kit with a sample. The present invention can easily and quickly detect a target substance by using fluorescence signal differences of fluorescent nucleobase analogues present in the split G-quadruplex DNA, and can be utilized for the detection of various biological materials and chemicals.
