The present invention relates to a method for simultaneously preparing a low-molecular functional biomaterial and a lactic acid bacteria nucleotide having utility in skin and cosmetics fields and, more specifically, to a method for preparing a lactic acid bacteria nucleotide (cubisome) inclusion-complexed to a low-molecular functional biomaterial, which is a 2-in-1 production method for simultaneously solving two problems of conversion of a high-molecular functional biomaterial to a low-molecular functional biomaterial and elution of lactic acid bacteria nucleotide components by a single cell crushing process. According to the present invention, a nucleotide derived from lactic acid bacteria, which effectively removes the intrinsic cytoplasmic protein and cell wall components of lactic acid bacteria affecting the production of fermentation odor of skin microbiome and raw materials and has increased stability by being inclusion-complexed with an exogenous low-molecular biomaterial, is produced at a high concentration, thereby being effectively used as a biomaterial in cosmetics, foods, and medicines.;COPYRIGHT KIPO 2020
