[技术领域]
本发明属于基因工程技术领域,具体地说涉及细胞色素P450 3A5的基因、含有该基因的的重组载体、用该载体转化的宿主、以及利用转化体生产的该基因的表达产物及其应用。
[背景技术]
细胞色素P450是重要的药物代谢I相酶,市场上80%以上的药物是通过细胞色素P450代谢的。3A亚家族是细胞色素P450基因家庭的主要成员,主要包括3A4、3A5及3A7几个成员。细胞色素P4503A5具有与3A4相似的代谢谱,能够代谢多种药物如midazolam,nifedipine及睾丸激素,孕酮等。
包括人、狗、大鼠、小鼠等多种动物的细胞色素P450基因序列已经被克隆,各种细胞色素P450也已经在大肠杆菌,酵母,哺乳动物细胞等宿主中表达。异源表达的细胞色素P450是研究药物代谢,进行体外药物筛选的重要手段。食蟹猴是常用的临床前安全性评价实验大动物,其与人的亲缘关系非常接近,因而在代谢上表现出较高的相似性。到目前为止,仍未见食蟹猴与人P450 3A5亚型相应的基因的克隆的报道。因此,克隆食蟹猴P450 3A5基因并进行异源表达对于体外药物筛选有重要意义。
P450酶在催化反应中需要两个电子和两个质子,其中的电子由内质网上的NADPH(还原型烟酰胺腺嘌呤二核苷酸磷酸盐,还原型辅酶II)-细胞色素P450氧化还原酶(NADPH-cytochrome P450oxidoreductae)提供。在体外反应中,缺少NADPH-细胞色素P450氧化还原酶的系统不具有催化活性。到目前为止,人及大鼠等的P450氧化还原酶基因已经被克隆,但仍未见食蟹猴P450氧化还原酶基因克隆的报道。
[发明内容]
本发明的目的在于克隆食蟹猴P450 3A5基因并在昆虫细胞中进行异源表达;还包括克隆食蟹猴NADPH-细胞色素P450氧化还原酶并在昆虫细胞中进行异源表达。
本发明通过以下方式达到克隆食蟹猴P450 3A5基因并进行异源表达的目的:根据已经发表的人P450基因序列(Genebank,AccessionNO.:NM_000777),以逆转录自食蟹猴肝RNA的cDNA为模板,通过聚合酶链式反应(PCR)扩增食蟹猴P450 3A5的编码区序列,扩增得到的PCR产物与pMD 18-T载体连接得到pT-cynoP3A5质粒。其中,设计的引物不包含除起始密码(ATG)外的食蟹猴P450 3A5的编码区序列。经测序,得到的编码区序列全长1512碱基对,编码503个氨基酸。
以上述克隆的食蟹猴P450 3A5的基因为模板,以包含RsrII及HindIII限制性内切酶位点的引物扩增食蟹猴P450 3A5的编码区序列并定向插入昆虫表达载体pFastBacl的RsrII及HindIII酶切位点之间,得到pFB-cynoP3A5表达载体。按照Bac-to-Bac BaculovirusExpression System Instruction Manual(由invitrogen公司提供)操作,在sf-9昆虫细胞中表达食蟹猴P450 3A5蛋白。异源表达的食蟹猴P450 3A5蛋白可以用作体外筛选,测试药物的代谢性质。
本发明通过以下方式达到克隆食蟹猴NADPH-P450氧化还原酶并进行异源表达的的目的:根据已经发表的人NADPH-P450氧化还原酶基因序列(Genebank,Accession NO.:NM_000941),以逆转录自食蟹猴肝RNA的cDNA为模板,通过聚合酶链式反应(PCR)扩增食蟹猴NADPH-P450氧化还原酶基因的编码区序列,扩增得到的PCR产物与pMD 18-T载体连接得到pT-cynoPOR质粒。。其中,设计的引物不包含除起始密码(ATG)外的食蟹猴P450 3A5的编码区序列。经测序,得到的编码区序列全长2034个碱基对,编码677个氨基酸。
以上述克隆的食蟹猴NADPH-P450氧化还原酶基因为模板,以包含RsrII及HindIII限制性内切酶位点的引物扩增食蟹猴NADPH-P450氧化还原酶的编码区序列并定向插入昆虫表达载体pFastBacl的RsrII及HindIII酶切位点之间,得到pFB-cynoPOR表达载体。按照Bac-to-Bac Baculovirus Expression System InstructionManual(由invitrogen公司提供)操作,在sf-9昆虫细胞中表达食蟹猴NADPH-P450氧化还原酶。异源表达的食蟹猴NADPH-P450氧化还原酶可以用作体外筛选,测试药物的代谢性质。
本发明的有益效果在于:分离的食蟹猴P450 3A5基因可以通过基因工程的手段在多种宿主中异源表达。异源表达的蛋白仅代表3A5这一亚型,因此其代谢活性也仅反映该亚型的活性。而食蟹猴肝及其亚细胞成分虽然亦含有P450 3A5蛋白,但同时也含有其他亚型的P450蛋白,因此它们的代谢活性不能代表单一的P450 3A5亚型的活性。
此外,本发明提供同是来源于食蟹猴的P450氧化还原酶,与使用其他种属的氧化还原酶构建的体外检测系统相比,用这一氧化还原酶构建的系统显然更接近于食蟹猴体内代谢的真实情况。

[具体实施方式]
1.sf-9昆虫细胞重组表达的食蟹猴P450 3A5的代谢咪哒唑仑活性测定
按技术方案所述,用制备的含有食蟹猴P450 3A5基因的杆状病毒以MOI(multiplicity of infection)=2感染培养到指数期的sf-9细胞,并在感染后2-3日收集细胞;
以0.1M磷酸盐缓冲液将收集的sf-9细胞匀浆;
以差速离心法分别在9000g,4℃离心20分钟及在100000g,4℃离心60分钟以制备微粒体;
将微粒体沉淀重悬,用BCA法测定蛋白含量,用一氧化碳结合法测定P450含量;
将上述微粒与系列稀释的咪哒唑仑混合物0.18mL置于反应管中,置37℃水浴保温5分钟。加20μL 10mM NADPH溶液启动反应。10分钟反应后加0.4mL含内标的反应终止液终止反应,混匀后置于冰水;
上述各反应管于2500g离心30分钟,取上清100μL加入到96孔板进样板,进行LC/MS/MS定量分析和数据处理,计算得到昆虫细胞重组表达的食蟹猴P450 3A5代谢咪哒唑仑的活性。
2.酮康唑(ketoconazole)体外抑制sf-9昆虫细胞重组表达的食蟹猴P450 3A5的活性测定
如实施例1中制备sf-9昆虫细胞重组表达的食蟹猴P450 3A5的微粒体;
将微粒体与固定浓度的睾酮以及系列稀释的酮康唑的混合物0.36mL置于反应管中,置37℃水浴保温5分钟。加40μL 10mM NADPH溶液启动反应。10分钟反应后加0.8mL含内标的反应终止液终止反应,混匀后置于冰水;
上述各反应管于2500g离心30分钟,取上清100μL加入到96孔板进样板,进行LC/MS/MS定量分析和数据处理,计算得到酮康唑抑制食蟹猴P450 3A5代谢睾酮的活性。
