Microarray Analysis in Support of Development of the Cynomolgus Macaque Model for Filoviruses.

This report details progress on the analysis of viral hemorrhagic fever virus infection by whole-genome microarray approaches. Over the past year we have received, prepared and run more than one hundred and ninety microarrays on samples received from USAMRIID. These arrays were from Ebola infected NHPs or Marburg infected NHPs. Following the collection of data on all of these arrays (in excess of two million data points) we have engaged in bioinformatics analysis to identify the significant changes seen in the circulating immune cells following virus infection. This has resulted in the assembly of a strong dataset for ebola virus infected animals and a more comprehensive analysis of Marbug virus infected animals. This work is the first systematic assembly of a picture of the whole-genome response to Marburg virus gene expression. This provides a significant cache of data to mine for early markers of infection, information on the nature of the immune response, and to use to identify novel targets of infection. Additionally, we have run deep-sequencing analysis of multiple samples of filoviruses which has allowed us to closely examine differences between virus strains.
