The distribution of a purified 125I-labeled alloantibody, pre-pared from the serum of Buffalo rats following immunization with an allogeneic Fischer tumor, was studied in normal Buffalo and Fischer rats. The immune Buffalo γ~globulin was labeled with 125I, and the 125I-antibody isolated by a twice repeated adsorption onto Fischer erythrocytes and elution from stroma prepared from these cells. Normal Buffalo γ-globulin was labeled with 131I. Five hours after intravenous injection of the paired labeled mixture into Buffalo rats, the labeled antibody was abundant only in plasma, behaving like normal γ-globulin. In contrast, in Fischer rats most of the label had left the plasma by 5.hours and become fixed predominantly in spleen, lung and liver.
