Elimination of Sugarcane mosaic virus and Sugarcane streak mosaic virus by tissue culture

Elimination of Sugarcane streak mosaic virus and Sugarcane mosaic virus from two commercial varieties of sugarcane viz. CoLk 8102 and CoJ 83 was attempted using in vitro culture technique. Plantlets were produced using meristem explants collected fromvirus infected field grown plants. RT-PCR assay of regenerated plants was carried out at monthly interval for six months using primer pairs for Sugarcane mosaic virus (SCMV-F3, SCMV-R3) and Sugarcane streak mosaic virus (SCSMV-S2, SCSMVPI). No virus could be detected in regenerated plants of both the varieties plantlets transferred to the glasshouse during the studies indicating the elimination of SCMVand SCSMV.
