Plant regeneration of Immature embryos of (Cariea papaya L.)

Studies were carried out on regeneration of plantlefs from immature embryos of papaya C4. SRS and CO.7 through embryo hesane. The embryos excised 50,60 and 70 DAP of both cv. SRS and CO.7 pre-chilled at 7 to 10 deg C for one day and cultured on full MS medium fortified with S.Omgh1. BAP and 1.0 mgr1 NAA, proved best for leaf emergence within a short period. The source plants (SRS and CO.7), which were gynodioecious forms, were sprayed with gentamycin 500 ppm, a week prior to as well as on the day ofcollection of explants to ensure sterile conditions. The immature fruits were collected 50, 60 and 70 DAP at regular intervals and washed with the detergent Tween 20' and then with distilled water. Finally, the fruit was cut open to extract the developing seeds along with sarcotesta. Since the embryos were located in the sterile environment of the ovule, surface sterilization of embryos was not necessary, instead, entire ovules were surface sterilized and then embryos were removed aseptically from the surrounding tissues. The process of excising immature embryos varies with species. However, many times, an incision can be made at the Mscropylar end of the young ovule and pressure applied at the opposite end to force the embryo out through the opening (Hu and Wang, (3).
