Pituitary adenylate cyclase-activating polypeptide (PACAP), a biologically active neuropeptide, has two amidated forms, PACAP-27 and -38, which are structurally similar to that of vasoactive intestinal polypeptide (VIP). Several studies reported that these peptides regulate gastric acid secretion but the results are not without controversy. In addition, their effects on pepsin secretion have been less explored. In the present study, we investigated the effect of PACAP on gastric acid and pepsin secretions in rats. Male SD rats were used after 18 h fasting. Under urethane anesthesia, chambered stomach was superfused with saline, and the acid secretion was measured by a pH-stat method. Pepsinogen secretion was measured by modified Anson method.
