Arrayed preparation of YAC DNA for pulsed field gel analysis

Measurement of insert size, as determined by, pulsed field gel electrophoresis (PFGE), is fundamental to many applications of yeast artificial chromosomes (YACs). Most available protocols for production of high molecular weight yeast eenomic DNA. suitable for PFGE, are designed for individual clone handling and laborious when applied to large numbers of YAC clones. MacMurray et al. have described a system for high throughput YAC DNA preparation in a %-weli format that can be used to produce materialsuitable for PFGE. Although largely automated, this method involves many liquid handling and transfer sieps with significant consumption of specialised plasticware, and is not suitable for laboratories lacking the appropriate robotic systems.
