Eight strains of bacteria isolated from soil under the heaps of cassava peels were screened for amylase production after growing on starch substrate. The various isolated strains were Corynebacterium spp (CoR,), Bacillus pumilus (BP,), Corynebacteriumspp (CoR2), Bacilus subtilis (BS,), Corynebacterium spp (CoR3), Corynebacterium bovis (CoR4), Corynebacterium bovis (CoRj), and Bacilus subtilis (BS2). Each of the strains gave an enhanced enzyme activity at pH 7.2 and a temperature of 65°C WxvBacilus subtilis (BS,) giving the highest activity 16.67. Oligosaccharide was produced by hydrolysis of cassava starch using amylase enzyme from Bacilus subtilis (BS,). Using Thin Layer Chromatography (TLC) plates with running solvent propanol : ethylacetate :water (11 : 2 : 7) and dinitrosalicyclic acid (DNS) as the locating reagent, maltose glucose, mannose, and fructose were obtained by reference to values obtained in the standard curve.
