Growth Kinetics Study of Free and Biofilm Cells of Avian Strain of Pasteurella multocida Field Isolate

An avian strain of Pasteurella multocida isolated from an outbreak in a poultry farm was grown in 3 and 0.32 per cent concentraions of tryptone soya broth (TSB) to obtain nutrient rich free cells (NRFC) and nutrient deficient free cells (NDFC), respectively and biofilm cells (BC) in 0.32 per cent TSB with 0.3 per cent bentonite clay to study the growth kinetics at different time intervals. Free cells (both NRFC and NDFC) reached peak growth on first day whereas BC on third day post inoculation. The culture in biofilm mode of growth was viable up to 51 days, compared to free cells which perished much earlier (NRFC and NDFC after 21 and 12 days, respectively). It was concluded that P. multocida cells with outer membrane protein biofilms survive significantly for longer period under in vitro environment (up to 51 days) compared to the free cells.
