In a preliminary experiment, 6 Beagles were injected i.v. with meloxicam at 0.2 mg/kg body weight or carprofen at 4 mg/kg, and blood samples were taken before and 10 and 30 min after injection. In a second experiment, the same dogs were given meloxicam at 0.05 or 0.1 mg/kg and ketoprofen at 0.5 mg/kg in a 3-way crossover experiment. The antiinflammatory agents were given orally once a day for 5 days, and blood samples taken before the start of treatment and 30 min after the last treatment. Inhibition of cyclooxygenase isoform 1 (COX-1) in platelets was measured indirectly by measuring the decrease in thromboxane B2 in whole blood ex vivo. The effects on COX-1 in kidneys were assessed by changes in urine volume and Na excretion. Meloxicam had a mild inhibitory effect and ketoprofen was a potent inhibitor of COX-1, whereas carprofen was intermediate.
