Words of wisdom: Re: In vitro production of functional sperm in cultured neonatal mouse testes.

The purpose of the recently published study was to evaluate the whole gametogenesis process in vitro using a murine model. The methodology used was the standard gas-liquid interphase method with two lines of transgenic mice and microinsemination. The authors demonstrated that the mei-osis progress in the cultured testis tissue, an alternative to fetal bovine serum known as knockout serum replacement, is more effective. In addition, the authors observed spermato-genesis in the peripheral region of each tissue piece, up to elongating spermatid formation. Additionally, the fertility of sperm cells produced in vitro was demonstrated by micro-insemination, and the reproductive status of the male and female offspring was proven. Furthermore, the authors demonstrated that fragments of the testis tissues can be cryo-preserved and can resume full spermatogenesis later in vitro.
