The aim of this study was to evaluate fibrolytic activity of rumen fungi by using disappearance of dry matter (DM) and neutral detergent fiber (NDF) and chitin content in rumen anaerobic fungi pure culture containing rice straw treated with sodium hydroxide (NaOH) for 12 days in in vitro condition. Treatments were: untreated rice straw and treated with NaOH (45, 70 and 120 g/ kg DM). The result showed disappearance of dry matter after 12 days incubation with rumen fungi will be 33.2, 38.3, 40.3 and 49.3 g/100 g DM for untreated rice straw and treated with 45, 70 and 120 g/kg DM NaOH, respectively. Sodium hydroxide caused to increase chitin content and disappearance of NDF and of media culture in compared with the other treatments, and the most increase of chitin content and NDF disappearance was for 120 g/kg NaOH, 12 days after culturing (168.6 mg/g DM and 5.96 g/kg DM, respectively) (P <0.05). It appears that the growth and activity of rumen fungi on rice straw are influenced by NaOH content.
