Traditionally, the detection of E. coli cells has been done using the colony counting method, which usually takes more than 24 hr. For many years, much effort has been made to develop a nonculturing method to detect these cells. This paper describes a rapid, sensitive, and simple enzymatic method for detecting E. coil cells. #beta#-galactosidase is a type of endoenzyme that exists in E. coli cells. It can be released from the cells by the pore-forming action of colicin El, and can be detected rapidly by enzymatic analysis with its substrate, o-nitrophenyl #beta#-D-galactopyranoside.
