Carnation is one of the most important flower crops in the world accounting for 17% of all cut flowers exported through the Dutch flower auction (Nugent et al., 14). In vitro micropropagation of carnation by using apical and axillary bud culture was reported (Roest and Bokelmann, 16; Mil et al., 10). Recent advances in the isolation, characterization and transfer of genes make genetic engineering an attractive technology to be used in conjunction with traditional breeding for the improvement of floricultural crops such as carnation (Woodson, 18). To establish a genetic transformation system, it is essential to develop an efficient shoot regeneration method. Adventitious shoot bud regeneration in carnation has been reported from hypocotyl (Petru andLanda, 15), shoot apices (Weryszko and Hempel, 19), axillary bud (Miller et al., 11, 12), stem (Nugent et al., 14) and leaf (Messfiguer et al., 9; Van Altvorst et al., 17) However, no detailed information has been available on the frequency of shoot-budregeneration. In the present investigation, an efficient system for adventitious shoot bud regeneration from stem and leaf derived calli of Dianthus caryophyllus cv Alas Red has been reported.
