Assessment of post-thaw seminal characteristics of Murrah buffalo bulls' spermatozoa by conventional and computer assisted semen analyser techniques

The present investigation was carried on 14 post-thaw seminal attributes of 110 collection corresponding to five different ejaculates from each of 22 Murrah buffalo bulls maintained at NDRI from the year 1987 to 1999 except 1996. Overall least squares mean of motility (percent), Live (percent), head abnormalities (percent), midpiece abnormalities (percent), tail abnormalities (percent), total abnormalities (percent), acrosome integrity (percent), hypo-osmotic reacted (HOS percent), CASA assessed motility (percent), VSL, (#mu#/sec), VCL (#mu#/sec), LIN (percent), ALH (#mu#) and VAP (#mu#/sec) were 44.45+-1.30, 63.21+-3.09, 7.12+-0.96, 2.75+-0.66, 10.73+-1.42, 19.67+-1.50, 49.48+-4.35, 36.52+-2.91, 47.25+-1.21, 29.58+-2.03, 84.40+-8.37, 41.54+-3.74,10.02+-1.44 and 54.59+-4.03, respectively. Analysis of variance revealed significant variation due to bull for head abnormalities (P < 0.01), acrosome integrity (P < 0.05), CASA assessed motility percent (P < 0.05), LIN (P < 0.01) and VAP (P < 0.05). Except for tail abnormalities (percent), all the post thaw characteristics were not affected significantly by season of freezing. Least squares analysis of variance could not reveal significant effect of year of freezing (storage interval). Significantly negative association of percent incidence of head and total sperm abnormalities with fertility performance (-0.484 and -0.531, respectively) suggested that initial screening of bulls based on these seminal characteristics could be helpful in further improving fertility performance.
