Multiple antipsychotic induced neutropenia - A possible genetic association

Clozapine-induced neutropenia and risk of agranulocytosis has resulted in well-formulated protocols (Berk et al., 2007). Neutropenia induced by other atypical antipsychotics, is less commonly reported. Mechanisms proposed include antipsychotic metabolite-induced neutrophil toxicity and immune system-mediated destruction (Class, 1989; Mason and Fisher, 1992; Flanagan and Dunk, 2008). A possible genetic basis is considered to contribute to the development of this idiosyncratic drug reaction (Carolin and Dettling, 2008). Studies since the early 1990s suggested that HLA haplotypes were associated with a patient's susceptibility to developing clozapine induced agranulocytosis (Dettling, 2001; Chowdhury et al., 2011).
