One of the major objectives of national and international research agenda in agriculture is to develop crop cultivars with improved tolerance to abiotic stresses such as drought, salinity and temperature extremes. For an efficient genetic transformation, organizing the genes and their respective promoters into a gene cassette is an art. Here we have developed a total of six gene constructs with three different cDNAs [Triticum aestivum Ascorbate peroxidase (TatAPX)]; Triticum aestivum C-repeat binding factor 2 (TaCBF2) and Arabidopsis thaliana C-repeat binding factor 3 (AtCBF3), driven by two different stress inducible promoters (AT LEA and RD29A). The six gene constructs were denoted as (i) pBI 221 PAtLEA:: TatAPX:: NOS, (ii) pBI 221 PAtRD29A:: TatAPX:: NOS, (iii) pDRIVE PAtLEA:: TaCBF 2:: NOS, (iv) pDRIVE PAtRD29A:: TaCBF2:: NOS, (v) pDRIVE PAtLEA:: AtCBF3:: NOS and (vi) pDRIVE PAtRD29A:: AtCBF3:: NOS. These gene constructs can be used in biolistic method of transformation to obtain transgenics for drought tolerance.
