Pigment as primary metabolites, and antibiotic as secondary metabolites can they form a relationship. Some pigmented antibiotic show strong correlation coefficient between antibiotic and intensity of culture colour. The relationship between antibiotics and pigments were statistically estimated. A total of 373 actinomycete isolates were investigated, 32 locally isolated actinomycetes were selected as they produce both pigment and antibiotic. These isolates were catalogued to 5 groups according to the nature of their culture soluble colour. All isolates have different action against test organisms. Methicillin-resistant Staphylococcus aureus (MRSA), Methicillin-sensitive S. aureus (MSSA), beta-Lactamase positive Escherichia coli and Candida albicanswere used as test organisms. Degree and type of soluble colour were identified according to Dictionary of Colour Standards (1951). Results revealed that blue pigment producer actinomycetes did not show strong relation as the blue pigment affected by thecarbon sources. Orange pigment group was affected by the elevation of temperature. Red, violet and yellow pigment producer's actinomycetes show strong correlation coefficient between pigment and antibiotic formation. Correlation coefficient (R2) value was 0.9154, 0.9754 and 0.9723 for isolate Or 3, V4 and Y7 respectively. These 3 isolates have shown the high correlation even when changing the substrate supplements.
