Backgrounds: The aim of this study was to investigate the effects of Gly-LDL on the proliferation, differentiation and expression of LRP5 and dickkopf-1 (DKK1) in mouse MC3T3-E1 cells. Methods: Mouse MC3T3-E1 cells were cultured with native LDL and various level of Gly-LDL for 48 h. Cell proliferation was measured by CCK8 assays, osteocalcin levels were determined by ELISA, and the expression of LRP5 mRNA and DKK1 mRNA was measured by real-time PCR Results: Gly-LDL inhibited MC3T3-E1 cell proliferation compared to that in the n-LDL control group. Calcein levels were decreased with Gly-LDL, but the expression of DKK1 mRNA was increased, and LRP5 mRNA decreased in MC3T3-E1 cells. Conclusions: Proliferation and differentiation were inhibited by Gly-LDL, which is related to the Wnt signaling pathway.
