Mesenchymal stem cells (MSCs) derived from bone marrow and umbilical cord Wharton's jelly have good potential for self-renewal and multi-linage differentiation. MSCs have been considered as an alternative extra-embryonic source for cell-based therapies. However, there is still little evidence comparing their proliferation and differentiation capacities. In this study, we extracted MSCs from rabbit bone marrow (BMSCs) and umbilical cord Wharton's jelly (WJ-MSCs) and determined their authenticity via the expression of surface antigens. Then, we compared the proliferation and differentiation capacity of MSCs at passage 4. QRT-PCR analysis revealed that WJ-MSCs expressed more CD90 than BMSCs, other than the CD73 and CD105. CCK-8, colony forming unit, and population doubling times assays did not show significant difference between BMSCs and WJ-MSCs. However, BMSCs expressed statistically more osteogenic, adipogenic and chondrogenic markers than WJ-MSCs. In conclusion, WJ-MSCs showed almost the same proliferation ability and sternness with BMSCs, while BMSCs showed greater osteogenic, adipogenic, and chondrogenic capacity than WJ-MSCs. Moreover, the optimal time for in vitro osteogenic induction before transplantation was 3 weeks, and the optimal time for both the adipogenic and chondrogenic induction was 2 weeks.
