Wharton's jelly (WJ) is a mucous connective extra-embryonic mesoderm used as a postnatal source for fetal stem cells. An in vitro 3-D Wharton's jelly scaffold made of human umbilical cord seeded with mesenchymal stem cells is described. Mesenchymal stem cells in scaffold (1 x 107 ml) were incubated in CO2, 37C and 90% humidity for 1h and 10ml media was added to each flask, incubated again for further cell attachment. Seeded samples were taken from freeze dried scaffolds sputter coated with gold for micro structural characterization and particle size analysis by scanning electron microscopy. We find that cells proliferated at different rates in scaffolds. Results suggest that the scaffolds appeared similar to 5 to 10 day egg-embryo divisions: by 15 days of culture, myocardiac muscle, fibrous and adipose tissue, and vascular blood vessels were present.
