The cultivated barley (Hordeum vulgare subsp. vulgare) is classified into two categories according to the adherence of caryopsis, hulled or naked barley. The hulled barley is often used for animal feeding and brewing industries, while the naked barley is desired for human food consumption. The naked trait was reported to be controlled by a 17-kb deletion harboring the entire ethylene response factor (ERF) gene on the nud locus that resides on the barley chromosome 7HL. To rapidly select the hulled and naked barley accessions, here we developed a new PCR-based diagnostic marker which produces smaller product with an improved resolution if relative to the former one. Considering that (1) there were recently reports revealed the existence of additional loci that control the adherence of caryopsis in cultivated barley, (2) the origin of naked genotype remains not yet conclusive, we applied the newly developed marker to carry out a screening of the nud locus among the globally-collected 1291 barley accessions. The genotyping result was fully coincidence with the caryopsis phenotype. 98.9% of naked barley accessions were derived from East Asia, and rare of accessions without hulls from other geographic places were found. Thus, our results suggested that the 17-kb deletion of barley ERF gene fully explained the compromised formation of adhesive caryopsis in this collection, and this trait can be selected with the newly-generated co-dominant diagnostic marker.
