Next generation sequencing. Third Generation Sequencer: New opportunities for de novo assembly

The PacBio RS (Pacific Biosciences) is the first Third Generation Sequencer and monitors the implementation of bases in real time. It is able to achieve sequencing reads much longer than any other technology. We adapted a workflow, in which PacBio RS data was corrected with Roche GS FLX reads. Sequencing of E. coli DH1 led to a lower amount of contigs in comparison to other sequencing approaches. The contigs were much longer accomplishing de novo assembly of small and mid-sized genomes more accurate.
