Assessment of Superior Individuals by RAPD in Zhongning Jujube Grown in Ningxia Hui Autonomous Region

摘要

In this paper, high quality genomic DNA were extracted from Zhongning jujube (Ziziphus jujuba Mill.) with an improved CTAB method. Seventeen primers were selected from 48 random ones to amplify 6 samples of Zhongning jujube including 4 desired individuals (P1-P4) and 2 normal ones (CK1, CK2). A total of 89 DNA fragments were obtained by using random amplified polymorphic DNA (RAPD) analysis with 10 random primers, and 89 of them were polymorphic bands. The genetic similarity coefficients were calculated by UPGMA cluster analysis. When D value was equal to 0.57, the 6 samples were classified into two clusters, one cluster included PI, P2, P3 and CK1, and another one contained P4 and CK2. The similarity coefficient between PI and CK2 was the minimum (0.333) showing they have farthest genetic relationship. Genetic difference between PI and P4 was much bigger than between P2 and P3. The results laid a foundation for further selection and breeding.
