A novel heparan sulfate disaccharide fluoride derivative of GlcN(SO3Na)α(l → 4)GlcAβ-F (1) was designed as a probe for detection of heparanase activity. Compound 1 was successfully prepared via 10-step chemical reactions. Human heparanase-1 effectively recognized 1, which induced liberation of the fluoride ion by hydrolysis via enzyme catalysis. The hydrolysis reaction was easily monitored by ~1HNMR measurements, in which the signal from H-1 gradually disappeared.
