Cryopreservation of red raspberry meristems (Rubus idaeus L.) isolated from in vitro plantlets

More economical maintenance of plant collections in vitro can be achieved through cryopreservation of the plant material, which abolishes the need to change nutrient media periodically. We studied the effect of sugars (sucrose, glucose) and cytokines (6-benzylaminopurine, thidiazuron) on the regeneration of apical meristems isolated from raspberry plants (Rubus idaeus L.) subjected to cold hardening in vitro and cryopreservation. Raspberry meristems, which retain their viability (varieties Skromnitsa and Lastochka), regenerated (10-86%) without entering the stage of calluses and yielded numerous progeny. It has been established that introduction of sucrose into the nutrient medium contributes to successful cryopreservation of raspberry apical meristems. We did not find any statistically significant differences between the effects of 6-BAP and thidiazuron on regeneration after cryopreservation of raspberry meristems.
