Solid-phase extraction and analysis of 5-fluoro-uracil plasmatic by high-performance liquid chromatography.5-Fluoro-uracil(5-FU)is an antitumoral agent widely employed in the treatement of many types of cancer.In the present study,a new reversed-phase high-performance liquid chromatography HPLC method has been developed and validated for the quantitative determination of 5-fluoro-uracil(5-FU)in human plasma.Before the chromatography,the drug(5-FU)was isolated from the plasma matrix by double extraction:liquid-and solid-phase extraction.Reversed-phase high-performance liquid chromatography was performed on a C18 column(250 mm x 4.6 mm I.D.)with a mobile phase of phosphate buffer(0.05 M,adjusted to pH 3.2)and ultraviolet detection UV(268 nm).
