Lateral buds excised from in vitro culture-derived shoots of A. arguta were subjected to pretreatments in freezing solutions, cold acclimation, and freezing in liquid nitrogen. A high survival rate was obtained after freezing and thawing in buds cold-acclimated at 5deg C for 4 weeks, followed by 0degree C for 2 weeks. 2. The following conditions optimal for the pretreatment of lateral buds were: (1) 15% DMSO in the freezing solution; (2) 1 h in the freezing solution; and (3) freezing solution kept at 0degree C. Half of the cold-acclimated lateral buds survived after treatment for 60 minutes in a freezing solution of 15% DMSO and 8 or 11.5 M glycerine, and prefreezing to -30degree C at 0.5degree C/minute before immersing in liquid nitrogen for 2 h.
