A new method based on dispersive liquid-liquid microextraction (DLLME) combined with high performance liquid chromatography (HPLC) was established for the determination of anthraquinone derivatives in human urine after oral administration of San-Huang tablets. Urine sample was pretreated with trichloroacetic acid, and then it was processed with DLLME. Some important parameters influencing the extraction efficiency, such as the type and volume of extraction solvent and dispersive solvent as well as the extraction time, were investigated in detail. Under the optimum conditions, the enrichment factors of aloe-emodin, rhein, emodin, chrysophanol, and physcion were 40, 93, 153, 131, and 119, respectively. Excellent linearity with correlation coefficient from 0.9996 to 0.9999 was observed for each anthraquinone derivative with limit of detection in the range of 0.15-2.56 ng·mL-1. Recoveries for the five anthraquinone derivatives were in the range of 81.9-96.6%. The established method has been successfully used to determine anthraquinone derivatives in human urine samples obtained from healthy volunteers following oral administration of San-Huang tablets.
