Plant regeneration protocol for Citrullus colocynthis L. was achieved from leaf and cotyledon explants. Explants were cultured on MS medium supplemented with 2,4-D, in combination with BAP, TDZ, Kn and Zeatin. Leaf explants are the best source for theinduction of high percentage of caulogenesis and regeneration (45%) on MS + 2.0 mg.Lr' Kn + 1.0 mg.IT1 TDZ. In cotyledon derived callus cultures 2,4-D + Zeatin combination played an important role in induction of multiple shoots (15-18) with 50% of regeneration response. Each cotyledon explants produce an average number of shoots (9.75 +- 0.2) with average shoot length 3.66 ±0.1.
