Sequence Analysis and Expression of One Fructose-Bisphosphate Aldolase Gene in Camellia oleifera

Fructose 1,6-bisphosphate aldolase, one of the improtant roles in the glycolysis/gluconeogenesis pathway, play significant regulatory roles in the processes of plant cell proliferation, growth, development, photosynthesis and stress resistance, etc. In this study, one fructose-bisphosphate aldolase gene full-length cDNA clone was isolated from the constructed Camellia oleifera cDNA library. Its ORF had 1179 base pair (bp) and designated as CoFBA1 (GenBank access numbers JN017093). It encoded an identical polypeptide of 393amino acids which had One fructose-bisphosphate aldolase class-I active site and three substrate binding sites. C. oleifera coFBA1 was classed into chloro-plast FBA and had 99% similarity with FBAs of dicotyledon and monocotyledon. The expression of CoFBA1 in seeds of three C. oleifera varieties and four growth periods was analyzed by real-time fluorescence quantitative PCR. The result showed 'Huashuo' had highest expression level, and in different growth periods the expression level decreased firstly and then increased. In conclusion, the cluster analysis with the observed expression levels indicates that they may play different roles in the processes of development, lipid biosynthesis and stress responses of C. oleifera seeds.
