Suspension calli of Satsuma mandarin (Citrus unshiu cv. Miyagawa Wase) were inoculated with Agrobacterium tumefaciens strain ERA 105 containing the binary vector pIG 121 harboring nptll and green fluorescent protein (GFP) genes, or pCAMBIA 1300 harboring hptll and GFP genes. In the selection of antibiotics, the calli were inoculated with Agrobacterium containing pIG 121 produced small amount of callus on 75 and 50 mg centry to L~(-1) kanamycin selection plates. The calli were inoculated with pCAMBIA1300 also produced callus clumps on 10 and 20 mg centry to L~(-1) hygromycin selection plates. GFP gene expression was observed in only three callus clumps produced on the 50 mg centry to L~(-1) kanamycin plate, but not in those on the hygromycin plate.
