Bone marrow examination gains increasing importance in veterinary medicine especially in the evaluation of patients clinically suspected of having hematological neoplasias. Microscopic examination of bone marrow smears however, is still very labourintensive and subject to observer variability. In the present study, we investigated the use of an automated hematology analyser, the ADVIA 120(R) multi species Hematology System (Bayer Diagnostics, Fernwald, Germany) for flow cytometric analysis of canine bone marrow. Bone marrow aspirates obtained from the iliac crest of 37 dogs referred to our clinic were collected in 10 ml-syringes prepared with K2-EDTA to prevent clotting. The samples were analysed by flow cytometry within two hours after collection and results were compared to those obtained by microscopic examination of May-Gruenwald Giemsa-stained bone marrow smears. Results of this study indicate, that the ADVIA 120(R) Hematology System provides a good screening tool for detection of lymphoproliferative and myeloproliferative disorders as well as for estimation of bone marrow cellularity and erythropoiesis. Due to flow cytometric analysis based on cytochemical staining and determination of cell lobularity, predominant cell lineages in bone marrow aspirates could be easily identified and were shown in characteristic ADVIA 120(R) cytograms.
