Glycerol is the essential cryoprotectant in all conventional freezing extenders for equine spermatozoa. It is used at concentrations from 2.5 to 6% (0.3-0.8 M). Several other cryoprotectants have been recently tested on stallion sperm. Among them, dimethyl formamide (at 0.6 M or 0.9 M) provides a similar protective effect on stallion spermatozoa to glycerol (at 0.55 M or 0.6 M) [1,2]. The aim of this study was to measure the effects of increasing concentrations of glycerol (G) and dimethyl formamide (DMF), added separately or in combination in the freezing extender, on the motility and the fertility of equine spermatozoa.
