Aim: The excretion in saliva of gemcitabine;its metabolite ','-difluorodeoxyuridine (dFdU) as well as epirubicin (Epi);its metabolite epirubicinol (Epi-ol) was studied in patients with non-small cell lung cancer;treated with gemcitabine plus epirubicin.Methods: Patients (n = ) were treated with gemcitabine mg/m;followed by Epi mg/m. Blood;saliva;oral mucosa cells were collected during hours for analysis of gemcitabine;Epi;their metabolites. Gemcitabine;dFdU;Epi;Epi-ol were quantified by high-performance liquid chromatography.Results: Gemcitabine was cleared rapidly from plasma;un-detectable after hours in all patients. Gemcitabine was detectable in saliva during only the first hour after infusion. The C_(max) in saliva was . +- . mg/L;the saliva to plasma ratio (S/P ratio) was . +- .. The C_max) of dFdU was reached .- hours after gemcitabine infusion;was . +- . mg/L. The dFdU S/P ratios gradually increased from . +- . at t = hour to . +- . at t = hours after infusion. Epi displayed a triex-ponential plasma concentration-time profile. The amount of Epi in mucosal cells ranged from - ng per cells at t - hours;decreased to - ng per cells at t = hours.Conclusion: Gemcitabine;Epi;as well as their main metabolites dFdU;Epi-ol;are excreted in detectable amounts in saliva;although their absolute concentrations remain relatively low.
