To evaluate the antioxidative activity of mackerel extracts, we examined the effect of mackerel extract on the production of glutathione (GSH) and reactive oxygen species (ROS) in HT-1080 cells. GSH levels were measured by using a fluorescent method employing monobromobiman (mBBr), and cellular oxidative stress caused by ROS generation from H2O2 was measured by performing the dichlorofluorescein-diacetate (DCFH-DA) assay. Treatments with acetone+methylene chloride (A+M) and methanol (MeOH) extracts both at 1 and 5 mg mL(-1) concentrations resulted in a significant increase in GSH levels compared with the control. The intracellular GSH levels also increased significantly in the presence of n-hexane and 85% aqueous MeOH fractions. The treatments with A+M and MeOH extracts (0.5 mg mL(-1) concentrations) inhibited H2O2-induced ROS production compared with the control without the extracts by 7.4% and 10.5%, respectively. Of all the fractions, the effect of 85% aqueous MeOH fraction was the strongest in reducing the concentration of ROS. Our results demonstrated that the 85% aqueous MeOH fraction inhibited cellular oxidation and that this fraction may contain valuable active compounds.
