Expression analysis of a bovine beta-casein/human lactoferrin hybrid gene in transgenic mice.

Milk was collected from mice with the transgene for human lactoferrin (hLF). Gene expression was analysed by Northern and Western blots, ELISA and immunohistochemical staining. Northern blot analysis of tissue from the mammary gland, liver, spleen,brain and pancreas showed that the transgene was expressed exclusively in the mammary gland. hLF was found in the milk from 5 of 8 transgenic mouse lines at concentrations of 2.5-200.0 鎔/ml. The apparent molecular weight of the recombinant hLF was indistinguishable from that of the native form upon immunoblotting. Immunohistochemical staining revealed that hLF in transgenic mice was exclusively expressed in the epithelial cells of the mammary gland.
