In vitro regeneration and transformation in chilli pepper (Capsicum annuum L.)

Regeneration of well-developed shootlets from cotyledonary explants of chilli (C. annuum) cultivars Pusa Jwala and G4 was obtained in Murashige and Skoog's (MS) medium supplemented with (9-18 #mu#M) zeatin in combination with GA3 [gibberellic acid] (2.89 #mu#M). The elongated shoots could be rooted successfully on IBA-containing MS medium. Cotyledonary explants of Pusa Jwala were transformed with Agrobacterium tumefaciens st C58 containing binary vector pGV1040 harbouring the two reporter genes npt II and GUS. The transformed shoots were selected on regeneration medium containing kanamycin (100 mg/litre) and cefotaxime (500 mg/litre) and subsequently rooted in the presence of kanamycin (75 mg/litre). The presence of the transgene was confirmed through histochemical staining of GUS, polymerase chain reaction and Southern hybridization analysis of npt II gene.
