Plant regeneration from somatic embryos of interspecific hybrids of grapevine formed in liquid medium.

Cell suspensions initiated from petiole-derived proembryogenic calluses gave rise to somatic embryos in three grapevine cultivars (Bianca, Intervitis Magaracha and Podarok Magaracha), all interspecific hybrids of Vitis vinifera and the American species V. riparia and V. rupestris. The cultivars differed in the subsequent behaviour of torpedo-stage embryos subcultured from liquid onto solid medium. Bianca produced shoots on hormone-free medium, while secondary embryos required medium with 0.5 mg benzyladenine (BA)/litre. In contrast, in Podarok Magaracha, shoots were produced on medium with BA, while secondary embryos were formed on hormone-free medium. In Intervitis Magaracha, secondary embryos were formed only on solid medium with 0.5 mg IAA/litre, and shoot development was observed on media with BA and on hormone-free medium. Secondary embryos were formed intensively on solid medium with 0.5 mg IAA/litre in all three cultivars. Plantlets with green cotyledons and hypocotyls which developed from torpedo-stage embryos in liquid medium with 0.5 mg BA/litre, and were subsequently cultured on solid, hormone-free medium, converted to callus. Germination of torpedo-stage embryos in liquid medium with 0.5 mg gibberellic acid (GA3)/litre for 5 days vs. 10 days incubation led to better subsequent shoot production from the developed plantlets with green cotyledons and hypocotyls after subculturing them onto solid medium with 0.5 mg BA/litre in all three grapevine cultivars..
