The applicability of PCR to detect Mycoplasma contamination in cell lines was investigated. 10 vero, 10 baby hamster kidney (BHK21) and 2 Madin Darby bovine kidney (MDBK) cell line samples were screened against Mycoplasma contamination. The DNA extraction from the sample, amplification of DNA and analysis of amplified DNA were done by slightly modifying the method described by van Kuppeveld et al. The presence of a band at 280 bp level was considered as a positive result for Mycoplasma contamination. Out of the 10 vero cell line samples tested, 7 were positive and 3 were negative. Out of 10 BHK21 samples tested, 3 were positive, whereas the rest were negative. All the MDBK cell line samples tested were negative for Mycoplasma contamination. These results indicated that PCR assay could be adopted for screening the cell line samples against Mycoplasma contamination.
