Effect of Different Antioxidants on Quality of Cryopreserved Boar Semen in Different Breeds

Cryopreservation is associated with the production of reactive oxygen species which lead to lipid peroxidation of sperm membranes that associated with reduction of sperm motility and decreased of fertilizing ability. The objectives of this study wereto examine the effects of 3 antioxidants supplementation to freezing extender II and freezing methods on post-thawed semen quality in different breeds of boars. Semen derived from 20 boars (Landrace (n=5), Duroc (n=5), Large White (n=5), and Pietrain (n=5)) of proven motility and morphology. Using a split-sample design, different antioxidants with X-cysteine 5 mM , glutathione 1 mM and water soluble Vitamin E 200 muM were supplemented. Each semen solution was splited into 2 groups for frozen with traditional nitrogen method and with controlled rate freezing method. After cryopreserved semen samples were thawed at 50 癈 for 12 second and were evaluated for progressive motility, sperm viability and acrosome integrity in live spermatozoa. The main findings emerging from this study were the supplementation of Z,-cysteine, glutathione, and water soluble Vitamin E to the freezing extender II resulted in an improvement of sperm motility, viability, and acrosome integrity of thawed spermatozoa. For freezing methods, controlled rate freezing method provided a significant superior in post-thawed semen qualities, compared with traditional nitrogen method. In addition, breeds of boars had also an effect on post-thawed semen qualities. In conclusion, supplementation of antioxidants directly into freezing extender II can improved post-thawed semen quality in different of boars, and freezing by controlled rate freezing method provided a higher post-thawed semen quality.
