Search for New Binding Sites for the Transcriptional Factor SF-1 by the SITECON Method: Experimental Verification and Analysis of Regulatory Regions of Orthologous Genes

As a result of analysis of 5' regions of genes involved in steroidogenesis by the SITECON method, 18 new SF-1 binding sites have been revealed (Table 2). Note that 15 out of 18 sites were discovered in the promoter regions of 32 genes, in which experimental search for SF-1 sites have not been performed so far (Table 2). In addition, three new potential sites were found in the genes that, according to TRRD data, contained experimentally identified SF-1 binding sites (Table 2): two in human and rat Cyp17 genes (positions -44 and -309) and one in the pig LHbeta gene (position -54). According to the estimation by the SITECON method, the level of conformational similarity of the SF-1 site at position -54 of the pig LHbeta gene is with the sequences of known SF-1 sites is lower than the value taken for recognition of all other sites. However, this site was also used for experimental verification, because its location was similar to the location of known SF-1 sites in orthologous genes (bull, horse, and rat LHbeta genes) (Fig. 2).
