Ourunderstandingofthemolecularmechanismsofprimordialgermcell(PGC)proliferationinfishisrudimentary,butitisthoughttobecontrolledbythesurroundingsomaticcells.WeassumedthatgrowthfactorsthatarespecificallyinvolvedinPGCproliferationareexpressedpredominantlyinthesurroundinggenitalridgesomaticcells.Inordertoisolatethesegrowthfactors,wecompiledacomplementaryDNA(cDNA)subtractivelibraryusingcDNAfromthegenitalridgesof40-dpfrainbowtroutembryosasthetesterandcDNAfromembryoswithoutgenitalridgesasthedriver.Thisapproachidentifiedanovelcytokine,designatedgonadalsoma-derivedgrowthfactor(GSDF),whichisamemberofthetransforminggrowthfactor(TGF)-βnbsp;superfamily.GSDFwasexpressedinthegenitalridgesomaticcellssurroundingthePGCsduringembryogenesis,andinboththegranulosaandSertolicellsatlaterstages.InhibitionofGSDFtranslationbyantisenseoligonucleotidessuppressedPGCproliferation.Moreover,isolatedtesticularcellsthatwereculturedwithrecombinantGSDFdemonstrateddose-dependentproliferationoftype-Aspermatogonia;thiseffectwascompletelyblockedbyantiserumagainstGSDF.TheseresultsdenotethatGSDF,anovelmemberoftheTGF-βnbsp;superfamily,playsanimportantroleforproliferationofPGCandspermatogonia./p/div
