Theexpressionandcoreproteinstructureoftwoproteoglycans,themajorcartilageproteoglycanisolatedfromaratchondrosarcomaandasmallmolecularweightchondroitinsulfateproteoglycanisolatedfromaratyolksactumor,havebeencompared.Thecartilageproteoglycanwasnotdetectableinthecartilagetissueofcartilagematrixdeficient(cmdcmd)neonatalmicebyimmunofluorescence,butthecmdcartilagedidreactwithantibodiesagainstthecoreproteinoftheyolksactumorproteoglycan.Radioimmunoassaysshowedthatthecoreproteinsoftheseproteoglycansarenotcross-reactivewitheachother.Analysisofthecoreproteinsbysodiumdodecylsulfate/polyacrylamidegelelectrophoresisafterchondroitinaseABCtreatmentoftheproteoglycanrevealedalargedifferenceintheirsizes.Thecartilageproteoglycancoreproteinhadamolecularweightofabout200,000whiletheyolksactumorproteoglycancoreproteinmigratedwithanapparentmolecularweightofabout20,000.Inaddition,theculturedyolksactumorcellsthatmakethesmallproteoglycandidnotreactwithantiserumagainstthecartilageproteoglycan.Theseresultsindicatethattheproteoglycanisolatedfromtheyolksactumorissimilartothesmallchondroitinsulfateproteoglycanspeciesfoundincartilageandsupporttheexistenceofatleasttwodissimilarandgeneticallyindependentchondroitinsulfateproteoglycancoreproteins.
