Manystepsofperipheralgliadevelopmentappeartoberegulatedbyneuregulin1(NRG1)signalingbuttheexactrolesofthedifferentNRG1isoformsintheseprocessesremaintobedetermined.Whileglialgrowthfactor2(GGF2),aNRG1typeIIisoform,isabletoinduceasatelliteglialfateinneuralcreststemcells,targetedmutationsinmicehaverevealedaprominentroleofNRG1typeIIIisoformsinsupportingsurvivalofSchwanncellsatearlydevelopmentalstages.Here,weinvestigatedtheroleofNRG1isoformsinthedifferentiationofSchwanncellsfromneuralcrest-derivedprogenitorcells.Inmultipotentcellsisolatedfromdorsalrootganglia,solubleNRG1isoformsdonotpromoteSchwanncellfeatures,whereassignalingbymembrane-associatedNRG1typeIIIinducestheexpressionoftheSchwanncellmarkersOct-6/SCIPandS100inneighboringcells,independentofsurvival.Thus,axon-boundNRG1mightactivelypromotebothSchwanncellsurvivalanddifferentiation.
