Neuralcrestcelldifferentiationisresponsivetoavarietyofextrinsicsignalsthatincludeextracellularmatrix(ECM)moleculesandgrowthfactors.Transforminggrowthfactor-?2(TGF-?2)hasdiverse,celltype-specificeffects,manyofwhichinvolveregulationofsynthesisofECMmoleculesandtheircellsurfacereceptors.WearestudyingbothseparateandpotentiallyinterrelatedinfluencesofECMandgrowthfactorsoncrestdifferentiationandreportherethatTGF-?2altersseveralaspectsofcrestcellbehaviorinvitro.Clustersofquailneuralcrestcellswereculturedinthepresenceandabsenceof400pMTGF-?21andexaminedat1,3,and5days.Whenexaminedat5days,therewasadramaticdecreaseinthenumberofmelanocytesintreatedcultures,regardlessoftheonsetordurationofTGF-?2treatment.WithcontinuousTGF-?2treatment,orwithtreatmentonlyduringcrestclusterformationonexplantedneuraltubes,manycellsincreasedinarea,becomingextremelyflat.ThesechangeswereevidentbeginningonDay3.Whilequantitativeanalysesofvideoimagesdocumentedthesizeincrease,severalaspectsofmotilitywererelativelyunchanged.Synthesisoffibronectin(FN)byapproximately11%ofcellsonDay3and31%ofcellsonDay5wasdemonstratedbyimmunocytochemistryandwasassociatedwithasixfoldincreaseinFNmRNAbyDay5.ExperimentswhichcorrelatedFNimmunoreactivitywithincorporationofbromodeoxyuridinesuggestedthatthepopulationoflarge,flat,FN-positivecellsdidnotproliferateselectivelyandthattherewasaslowerrateofproliferationinTGF-?2-treatedculturesthaninuntreatedcultures.ThelargeFN-immunoreactivecellsresemblecellsderivedfromcepalicneuralcrestandraiseinterestingquestionsconcerningpotentialrolesforTGF-?2inregulatingcrestdifferentiationinvivo.
