Whena13-day-oldchickembryonictarsometatarsalskinwasculturedfor4daysinmediumcontaininghydrocortisone(20nM)and5%delipidizedfetalcalfserum(FCS),epidermalgrowthfactor(EGF,100ng/ml)decreasedepidermalDNAcontent42%andinhibitedepidermalDNAsynthesis87%.TonofilamentbundleswithinthebasalandintermediatecellsoftheEGF-treatedepidermiswerenotasconspicuousasthoseintheglucocorticoid-inducedkeratinizedepidermis,andtheupperregionoftheEGF-treatedepidermisdidnotformeitherthefilamentbundlesortheelectron-denseamorphousmassesseeninthecytoplasmoftheglucocorticoid-inducedkeratinizedlayer.EGFstimulateddegradationofglucocorticoid-induced?±-keratin.Furthermore,EGFcausedatwofoldincreaseinglucocorticoid-inducedepidermaltransglutaminaseactivityandintheamountofepidermalglucocorticoidreceptor.IntheabsenceofFCS,however,EGFdidnotinhibitsteroid-induced?±-typekeratinizationanddidnotaffecteithersteroid-inducedepidermaltransglutaminaseactivityoramountofepidermalglucocorticoidreceptor.Hence,theeffectofEGFonglucocorticoid-inducedepidermaltransglutaminaseactivitywasobservedonlyinthepresenceofdelipidizedFCSandmightbesupportedbyanincreaseintheamountofglucocorticoidreceptor.
