Adulttissuesoftheseaurchin,Strongylocentrotuspurpuratus,wereanalyzedfortheproductsofasetofgeneswhoseexpression,intheembryo,isrestrictedtotheskeletogenicprimarymesenchyme(PM).ThreeembryonicPM-specificmRNAswerefoundtobeabundantinadultskeletaltissues(testandlantern),butnotinavarietyofsofttissues.HomologousmRNAswerealsofoundinskeletaltissuesofthecongenericseaurchin,S.droebachiensis,aswellasamoredistantlyrelatedechinoid,Dendrasterexcentricus,andanasteroid,Evasteriastroschellii.ThedistributionsoftwooftheseRNAswereanalyzedinregeneratingspinesofadultS.purpuratususinginsituhybridization.Thesegeneproductswerelocalizedprimarilyinthecalcoblaststhataccumulatedattheregenerationsite.InnonregeneratingspinesSpLM18RNAs,themostabundantofthesegeneproducts,werelocalizedinasmallpopulationofnoncalcoblastcellsscatteredthroughthespineshaft,andwereabsentfromcalcoblasts.Theseobservationssuggestthataprogramofgeneexpressionassociatedwiththeprocessofcalcificationisconservedbothdevelopmentallythroughtheperiodofmetamorphosisandevolutionarilyamongtheechinoderms.
